概要: The changes in proteoglycan (PG) expression and glycosaminoglycan (GAG) constituents in the intervillous space of the pregnancy-induced hypertension (PIR) placenta were investigated. PGs and GAGs were purified from the extract of the placental intervillous space by the DEAE-Sephacel column and salt-concentration gradient method. and the GAG sugar chains were released by the actinase and cellulase treatments. The sugar chains from the placentas of normal pregnancy and PIR were compared by cellulose-acetate membrane electrophoresis. No difference was observed in the expressions of hyaluronic acid. heparan sulfate, and chondroitin sulfate. but a clear increase in the expression of dermatan sulfate (DS) in the placenta of the PIR was confirmed. An increase of the DS that specifically activates anticoagulants can be a body reaction to counteract the hemostatic condition observed in PIR. 続きを見る

概要: The aim of study is to analyze functional sugar chains, glycosaminoglycans (GAGs) in urine frompostmenopausal women with osteoporosis. A 24-hour urine was collected from 15 osteoporotic women and normalwomen. All women were postmenopausal and aged at 60 to 70 years old. Diagnosis of osteoporosis was based onthe diagnosis criteria proposed by the Japan Osteoporosis Society after measurement of lumbar spine bone mineraldensity by dual energy X-ray absorptiometry. The urine was concentrated by dialysis and evaporation, thenprecipitated with ethanol. The precipitate was then reconstituted into NaCl solution, precipitated again by adding 10% cetylpyridinium chloride and the precipitation was used as complex GAGs fraction. The yield of complex fractionwas significantly higher in osteoporotic subjects than that in normal control. Then, the complex GAGs fraction wasanalyzed by cellulose acetate membrane electrophoresis and high performance liquid chromatography (HPLC),respectively. Cellulose acetate membrane electrophoresis has revealed that GAGs in urine contained partiallydesulfated chondroitin sulfate. However, further analysis by HPLC has shown that the excretion of desulfated GAGswas rather less in osteoporotic women, indicating that urinary GAGs fraction from osteoporotic women containsunknown acidic polysaccharides as well as GAGs. 続きを見る

概要: Decorin, a component of the low-molecular-weight proteoglycans (PG) of yellow ligaments was purified, and the structures of the core protein and the glycosaminoglycan (GAG) chains were analyzed. The crude PG fraction extracted under dissociative conditions was subjected to DEAE-Sephacel ion-exchange chromatography, gel-filtration on Sepharose CL-4B, and Octyl-Sepharose CL-4B chromatography, and decorin was obtained. The amino-terminal amino acid sequence of decorin was found to be identical to those of human bone PG-II and human fetal membrane PG-II. The analyses of the GAG chains using two-dimensional electrophoresis on cellulose acetate membranes, affinity high-performance liquid chromatography (HPLC) with a hydroxyapatite column and gel-filtration HPLC after chondroitinase digestion, showed the presence of dermatan sulfate (DS) chains and chondroitin sulfate (CS).
